With the recent addition of tylvalosin, our portfolio now includes an extra macrolide alongside tylosin and tilmicosin. This is an excellent opportunity to take a closer look at macrolides and provide an overview of the similarities and differences between these products. In this article, we focus specifically on their use in swine.<\/strong><\/p>\n

Macrolides \u2013 A Quick Refresher<\/strong><\/h3>\n

Depending on the active substance, macrolides are narrow- to broad-spectrum antibiotics and are generally classified as bacteriostatic. Several factors\u2014such as concentration and the bacterial strain\u2014can determine whether their effect becomes bactericidal. Their activity is based on inhibition of bacterial protein synthesis. To achieve this, the molecule must enter the bacterial cell and bind to the ribosomes (specifically the 50S subunit). This binding prevents amino acids from linking together to form a complete peptide chain. In the presence of macrolides, only incomplete protein chains are formed, ultimately leading to growth inhibition or bacterial death.<\/p>\n

Both aerobic and anaerobic Gram-positive bacteria and a limited number of Gram-negative species are susceptible. After rapid absorption from the gastrointestinal tract or injection site, macrolides are widely distributed throughout the tissues. Tissue concentrations often exceed serum concentrations many times over. This is due to their basic chemical nature and their tendency to accumulate in acidic environments. Excretion occurs mainly via the faeces.<\/p>\n

Unfortunately, resistance to macrolides can develop relatively quickly, and cross-resistance may also occur.<\/p>\n

Comparing Oral Administration of Tylosin, Tylvalosin and Tilmicosin<\/strong><\/h3>\n

Dopharma\u2019s range includes oral formulations of tylosin (Tylogran), tylvalosin (Aivlosin) and tilmicosin (Tildosin). While tylosin is a natural product produced by Streptomyces fradiae<\/em>, tylvalosin and tilmicosin are semi\u2011synthetic derivatives of tylosin, originally developed in part to overcome emerging tylosin resistance.<\/p>\n

As noted earlier, oral administration results in rapid absorption, with peak blood levels observed within minutes to hours. Distribution to various tissues is equally fast and effective. In tissues, tilmicosin maintains a prolonged high therapeutic concentration, whereas concentrations of tylosin and tylvalosin decline significantly more quickly. As a result, tylosin and tylvalosin have considerably shorter withdrawal periods.<\/p>\n

All three products share an indication for respiratory disease, targeting Mycoplasma hyopneumoniae<\/em>. Tylvalosin is particularly focused on this pathogen, and Aivlosin has even been associated with potential eradication of M. hyopneumoniae<\/em> in certain contexts. Tilmicosin, however, has an even broader spectrum within respiratory conditions, as it is also effective against Pasteurella multocida<\/em> and Actinobacillus pleuropneumoniae<\/em>. Furthermore, tilmicosin has been shown in vitro to reduce replication of the Porcine Reproductive and Respiratory Syndrome (PRRS) virus in alveolar macrophages in a dose-dependent manner.<\/p>\n

Tylosin and tylvalosin are both effective against Lawsonia intracellularis<\/em> (PIA), with tylosin having a long-established track record in this area.<\/p>\n
